SIRPα(Signal Regulatory Protein α)是一種免疫球蛋白超家族跨膜蛋白,主要表達于 髓系細胞(如巨噬細胞、樹突狀細胞)表面,其胞外區含三個 Ig 樣結構域(D1、D2、 D3),其中 D1 結構域特異性結合 CD47;胞內段含免疫受體抑制基序(ITIM), 可招募 SHP-1/SHP-2 磷酸酶傳遞抑制信號。SIRPα通過調控吞噬活性維持自身免疫耐受, 防止巨噬細胞誤傷正常細胞。 CD47 是一種五次跨膜蛋白,廣泛表達于紅細胞、血小板及腫瘤細胞表面,其胞外 N 端 IgV 樣結構域與 SIRPα結合,被稱為“別吃我"(Don’t eat me)信號分子,在紅細胞穩 態(防止脾臟巨噬細胞清除年輕紅細胞)及腫瘤免疫逃逸中起關鍵作用。 當 CD47 結合 SIRPα后,SIRPα胞內 ITIM 基序被磷酸化,招募 SHP-1/SHP-2 磷酸酶, 通過去磷酸化作用抑制下游促吞噬信號。 | |
